| description | Bioethanol plants face challenges in the maintenance of using production waste, such as stillage. For every 1 liter of bioethanol, 10-15 liters of stillage are generated. It can be used as a raw material for the production of concentrated feed, or stillage, which is subjected to methane fermentation to obtain a gaseous energy carrier - biogas. In this way, this Thus, the production waste is valorized, and there is a the possibility of replacing fossil fuels in the production of bioethanol appears. The commissioning of industrial biogas reactors is associated with certain difficulties. Our task was to generate a model simulate the commissioning of a reactor start-up for biogas fermentation of stillage, which comes from an operating plant with a capacity of 170 m3 of bioethanol per day. To carry out the biogas fermentation process, a laboratory reactor with a working volume of 300 l was used, equipped with a temperature stabilization system (electric heating), an electric mixer, devices for periodic loading of the substrate and removal of the fermented liquid, a gas removal and accounting system using a meter. Methane fermentation was controlled by FOS/TAC indicators. The substrate (stillage) was taken directly from the plant, with a stable technological mode of bioethanol production throughout the experiments. The inoculum was digestate (a by-product of biogas production) collected from an operating industrial biogas plant. During the work, the composition of the raw material and inoculum was studied, and the pH and temperature of the biogas process were controlled. The methanogenesis process took place under thermophilic conditions, at 45-50 °C, the substrate load on dry matter was 4 ± 0.4 kg/m3 per day. The specific biogas yield during the experiment was 48.5±1.0 m3 per m3 of bard. The methane content in biogas during the period of stable fermentation (13…25 days) was 60.7 ± 0.8% by volume. Based on the results of the experiments, it can be concluded that the addition of lignocellulosic substrate will increase the biogas yield. Given the level of FOS/TAC, when commissioning and operating an industrial reactor for wort fermentation, the organic matter loading can be recommended to 5…6 kg/m3 per day. The data obtained will be used when commissioning reactors of an industrial biogas plant for stillage processing in an amount of 1000…1100 m3 per day. |

МОДЕЛЮВАННЯ ПОЧАТКОВОЇ СТАДІЇ МЕТАНОВОЇ ФЕРМЕНТАЦІЇ ЗЕРНОВОЇ БАРДИ
В ЛАБОРАТОРНОМУ РЕАКТОРІ

Анотація. На біоетанольних заводах існує проблема з
утилізацією такого відходу виробництва, як барда. На
кожний 1 л біоетанолу утворюється 10–5 л барди. Її мо-
жна використовувати як сировину для виробництва
концентрованих кормів або піддавати метановій фер-
ментації для отримання газоподібного енергоносія – бі-
огазу. У такий спосіб відбувається валоризація цього
відходу виробництва, і є можливість заміщення викопних енергоносіїв для виробництва біоетанолу.
Введення в дію промислових біогазових реакторів пов’язане з певними складностями. Нашою задачею
було моделювання введення в дію реактора для біогазової ферментації барди, що виходить з діючого
заводу потужністю 170 м3 біоетанолу на добу. Для проведення процесу біогазової ферментації викори-
стовували лабораторний реактор, робочим об’ємом 300 л, обладнаний системою стабілізації темпе-
ратури (електричний підігрів), мішалкою з електроприводом, пристосуваннями для періодичного зава-
нтаження субстрату й виводу збродженої рідини, системою виведення й обліку газу за допомогою
лічильника. Контроль метанової ферментації здійснювали за показниками FOS/TAC. Субстрат (барда)
відбирався безпосередньо з заводу за стабільного технологічного режиму виробництва біоетанолу
протягом періоду виконання експериментів. Як інокулят слугував дигестат (побічна речовина після
отримання біогазу), який був відібраний з діючої промислової біогазової установки. У процесі роботи
досліджувався склад сировини та інокуляту, контролювалися рН та температура біогазового процесу.
Процес метаногенезу відбувався у термофільних умовах, при 45–50 оС, навантаження субстратом за
сухою речовиною 4 ± 0,4 кг/м3 на добу. Питомий вихід біогазу під час експерименту становив 48,5 ± 1,0
м3 на кожен кубічний метр барди. Вміст метану в біогазі в період стабільної ферментації (13…25 діб)
становив 60,7 ± 0,8 % об’єму. Виходячи з результатів експериментів, можна зробити висновок, що до-
давання лігноцелюлозного субстрату підвищить вихід біогазу. З огляду на рівень FOS/TAС під час вве-
дення в дію і експлуатації промислового реактора для ферментації барди навантаження органічною
речовиною можна рекомендувати до 5…6 кг/м3 на добу. Отримані дані будуть використані при введенні
в експлуатацію реакторів промислової біогазової установки для перероблення барди в кількості 45…55
м3 на добу.

Огляд, проблематика
Біоетанол в значній кількості виробляється з крохмале-
вмісної сировини – зерна кукурудзи, ячменю, жита,
пшениці [1, 2]. Насамперед це некондиційне і дефектне
зерно кукурудзи й інших злаків, відходи зернопереро-
бки, які непридатні для харчових або кормових цілей.
Сировина піддається термоферментативній обробці, і
отриманий розчин цукрів ферментується дріжджами в
етанол [3].
Побічний продукт після відокремлення етанолу від куль-
туральної рідини має назву «післяспиртова зернова
барда», далі «барда». Барда містить усі складові компо-
ненти вихідної сировини, за винятком крохмалю й неве-
ликої частки інших вуглеводів, що перетворились дріж-
джами на етанол. Якщо якість крохмалевмісної сировини
задовільна, барда може служити сировиною для вироб-
ництва продуктів кормового призначення [4, 5].
Існує ще один шлях утилізації барди – її анаеробна фер-
ментація з метою отримання біогазу [6–8]. Останнім ча-
сом дедалі більше біоетанольних підприємств викорис-
товує дешеві відходи й дефектну крохмалевмісну
сировину, які не відповідають ветеринарним вимогам і

можуть містити токсини, що унеможливлюють викорис-
тання барди для виробництва кормів [9]. У цьому разі
післяспиртову барду доцільно піддавати метановій фе-
рментації для отримання газоподібного енергоносія –
біогазу або біометану [10]. У такий спосіб відбувається
валоризація небезпечного відходу виробництва і част-
кове заміщення викопних енергоносіїв для виробниц-
тва біоетанолу [11].
1. Постановка задачі
Як субстрат для біогазової ферментації (Anaerobic
Digestion – AD) барда має свої особливості, що треба
брати до уваги під час проєктування і введення в
експлуатацію промислових біогазових установок. Зага-
лом барда відрізняється від традиційних субстратів
для AD, як-от відходи тваринництва і лігноцелюлозна
біомаса, відносно низьким співвідношенням вуглецю
та азоту (С/N) і більш низьким значенням рН. Вона
може бути піддана AD при досить високому наванта-
женні за органічною речовиною (Organic Loading –
OL) – до 10 кг на 1 м3 реактора за добу і гідравлічному
часі перебування в реакторі (Hydraulic Retention Time –
HRT) від 6 до 40 діб без вимивання метаноутворюваль-
ної мікрофлори. Але за високих навантажень зни-
жується питома кількість біогазу (Specific Biogas
Production – SBP), що утворюється від органічної речо-
вини, здатної до біоконверсії (Volatile Solids – VS), – з
650 до 550 L kg−1 [12].
Крім того, в барді порівняно мало природних полімер-
них сполук (наприклад, целюлози). Ці особливості, оче-
видно, мають вплив на протікання мікробіологічних
процесів AD барди, що приводять до утворення біогазу.
Через те що співвідношення карбону до нітрогену (С/N)
є занизьким для біогазової ферментації, та з огляду на
високий вміст калію, сульфатів та фенольних сполук, до
барди часто треба додавати косубстрати. Проблема
може вирішуватись додаванням лігноцелюлозної біо-
маси. Для цього придатні різноманітні відходи агропро-
мислового комплексу, які містять целюлозу та інші ву-
глеводи [13, 14].
Відомо, що введення в дію біогазових реакторів
пов’язане з певними ризиками. Початок процесу AD
може бути досить тривалим через несприятливі зміни
рН внаслідок неконтрольованого зростання концентра-
ції летких кислот жирного ряду (ЛЖК), сполук амонію
або негативний вплив сполук сірки тощо. Надмірне
накопичення ЛЖК може спричинити «закисання» сере-
довища, тобто неприпустиме зниження рН. Навпаки,
відносно високий вміст амонійних сполук у реакторі
може привести до перевищення оптимальних для мета-
ногенезу значень рН, особливо при термофільному ре-
жимі.
Підприємство, що виробляє 170 м3 біоетанолу на добу з
крохмалевмісної сировини, використовує технологію
Very High Gravity (VHG) ферментації [15]. Ще одною
особливістю технології, яка дає нетиповий склад зерно-
вої барди, є використання для етанольної ферментації
комбінованої сировини – зернового сорго, крохма-
левмісних понад 17 % v/v [16]. При цьому барда теж має
підвищену концентрацію сухих речовин (CP >12 % w/v) і
ЛЖК (переважно оцтової кислоти – до 0,5 % v/v.). Пла-
нується будівництво біогазової установки для утилізації
барди з отриманням біогазу.
Метою роботи було визначення умов, за яких можна
здійснити початкову стадію процесу і виходу анаероб-
ного реактора на робочий термофільний режим при ви-
користанні моносубстрату – зернової барди підвищеної
щільності.
Поставлена задача. Змоделювати введення в дію ре-
актора для AD барди в термофільних умовах. Визна-
чити період виходу реактора на стабільний режим ро-
боти при навантаженні субстратом за сухою
речовиною 4 ± 0,4 кг/м3 на добу. Отримати показники
й межі їх змін, за якими буде здійснюватися контроль
процесу, оцінити питомий вихід і якість біогазу.
Оцінити перспективу підвищення навантаження реак-
тора за сухою речовиною. Визначити зміну співвідно-
шення Карбон / Нітроген у процесі AD і можливість
підвищення продуктивності реактора за рахунок ви-
користання додаткових субстратів з високим
співвідношенням С/N.
2. Матеріали і методи
Для проведення процесу біогазової ферментації вико-
ристовували лабораторний СSTR-реактор з об’ємом
робочої частини 300 л, обладнаний системою стабілі-
зації температури (електричний підігрів, контрольова-
ний за показником температури середовища), мішал-
кою з електроприводом, пристосуваннями для
періодичного завантаження субстрату й виводу збро-
дженої рідини, системою виведення й обліку газу за
допомогою лічильника ГБР400 (схема і фото реактора
зображені на рис. 1).
Склад газу (СН4; СО2; Н2S) визначали автоматичним пор-
тативним цифровим газоаналізатором марки Landtec
GEM-500 при лабораторному дослідженні та за допомо-
гою газового хроматографа Aglient 7890B GC System
(США). Аналізи субстрату й культуральної рідини вико-
нували в лабораторії заводу за відповідними стандарти-
зованими методиками.
Контроль метанової ферментації здійснювали за показ-
никами FOS/TAC. Показник FOS (нім. Flüchtigen
Organischen Säuren) є кількісною мірою летких органіч-
них кислот, а TAC (англ. Total Anorganic Carbon) є мірою
лужної буферної ємності [17, 18].
Крім того, для стабільного процесу біогазової фермен-
тації дуже важливо було фіксувати також температуру в
реакторі, температуру вхідної сировини, вміст сухих ре-
човин (СР), особливо органічних сухих речовин (ОСР), і
рН середовища в реакторі протягом всього процесу ана-
еробної ферментації.

Субстрат для біогазової ферментації (барда) відбира-
вся безпосередньо з заводу при стабільному техноло-
гічному режимі виробництва біоетанолу протягом пе-
ріоду виконання експериментів. У технологічному
процесі виробництва біоетанолу для стабілізації рН фе-
рментаційного середовища традиційно використову-
ється сірчана кислота. Для запобігання можливому
надмірному утворенню сірководню при метановій фе-
рментації в технологічному процесі виробництва біое-
танолу протягом одного тижня замінили сірчану кис-
лоту молочною кислотою. У табл. 1 наведені резуль-
тати аналізу складу барди при використанні сірчаної та
молочної кислоти. Для аналізу відібрані середньодо-
бові проби при використанні сірчаної й молочної кис-
лот за умов, що унеможливлювали змішування зразків
з різними кислотами.
Методики вимірювань – відповідно до Державних стан-
дартів України (ДСТУ) та Методик виконання вимірю-
вань (МВВ 18/01-2019).
3. Аналіз сировини
Найважливіші показники барди, яка використовувалась
у експериментах, наведені у табл. 1.

Судячи з отриманих даних, вміст сірки та інших компо-
нентів у зразках барди з використанням різних кислот
відрізняється незначно, близько до розширеної неви-
значеності. Тому під час експериментів з анаеробної фе-
рментації було прийняте рішення не брати зазначені
зміни складу субстрату до уваги. Експерименти викону-
вали з субстратом (бардою), який надходив з діючого
підприємства, незважаючи на те, яка кислота викорис-
товувалась у технологічному процесі.
Перед початком експериментів лабораторний реактор
завантажили інокулятом, яким слугував дигестат (побі-
чна речовина після отримання біогазу), що був відібра-
ний з діючої промислової біогазової установки. Фізико-
хімічні показники інокуляту наведені в табл. 2.

4. Опис експерименту
Інокулят для початку ферментації був відібраний з дію-
чої промислової біогазової установки, в якій у мезофіль-
ному режимі (32…36 оС) перероблявся комплексний
субстрат з нестабільним співвідношенням компо-
нентів – відходи лігноцелюлозної біомаси, гній ВРХ, бу-
ряковий жом тощо. Дигестат з цієї установки планується
використати як інокулят для введення в дію установки з
переробки зернової барди. Тому лабораторне моделю-
вання процесу було виконане саме з використанням за-
значеного інокуляту. Щоб запобігти тепловому стресу
мезофільної мікрофлори інокуляту, процес АD в лабора-
торному реакторі починали з мезофільного режиму.
Дигестат з вищезгаданої діючої промислової установки
(інокулят), доставлений для виконання експерименту,
мав температуру близько 24 оС. Лабораторний реактор
заповнили інокулятом у кількості 300 л і довели темпе-
ратуру рідини в реакторі до 27,3 оС. Після цього
завантажили 10 л барди і включили стабілізацію темпе-
ратури на рівні 35 ± 0,5 оС. Подальше завантаження
здійснювали також по 10 л барди один раз на добу з
приблизного розрахунку навантаження 4 г органічної
сухої речовини на 1 дм3 об’єму реактора і часу перебу-
вання рідини 30 діб. При завантаженні відбирали
відповідний об’єм дигестату для підтримання сталого
об’єму рідини в реакторі.
Температуру в лабораторному реакторі підіймали при-
близно на 2 оС кожної доби до досягнення 40 оС, а далі –
на 1 оС кожної доби до досягнення 45 оС, і на 0,5 оС до
досягнення ≈50 оС. У такий спосіб моделювали процес
введення в дію промислової біогазової установки при
виведенні її на термофільний режим.
Результати та обговорення
Дані аналізів та вимірювань протягом всього періоду
експерименту зведені в табл. 3.

Вихід на запланований рівень температури в реакторі
здійснювали протягом 14 діб (див. табл. 1). У цей період
проходила адаптація мікроорганізмів до підвищення
температури й до навантаження субстратом. При цьому
вміст летких кислот (FOS) зростав, і міг відбутися вихід
середовища за межі оптимального діапазону рН 7…8. У
цьому разі потрібно було б знизити швидкість підйому
температури та/або завантаження субстрату через пе-
ревищення адаптивної спроможності мікрофлори в ре-
акторі.
У цей період особливу увагу приділяли зміні показника
FOS/TAС у реакторі, слідкуючи за тим, щоб вміст летких
кислот (FOS) відносно лужності (TAС) середовища не пе-
ревищував допустимого значення – 0,5. Тому періодич-
ність завантаження субстратом і його кількість не
змінювали.
З 15-ї по 27-му добу процес метагенезу вийшов на
стабільний режим. Лужність (TAС) інокуляту і субстрату
була стабільною протягом усього експерименту завдяки
сталому вмісту карбонатів у барді. Порівняно низький
вміст амонійного азоту в барді не спричиняв надмірного
підняття рН у реакторі як це відбувається, наприклад, у
разі перероблення субстратів з вмістом курячого посліду
[19].
Показник FOS/TAС після закінчення періоду адаптації
був у межах стабільного процесу – 0,3…0,4.
Інокулят мав початкову концентрацію 1,3 % СР, тому в
період адаптації цей показник зростав до встановлення
балансу між надходженням речовини в реактор і вихо-
дом її з біогазом. Коливання вмісту СР у реакторі після
стабілізації процесу пояснюється наявністю осаду й по-
траплянням його в проби рідини. Вихід біогазу протягом
експерименту показаний на рис. 3. Загалом динаміка
утворення біогазу і зміна його складу (рис. 4) була в
очікуваних межах, зважаючи на стабільний склад суб-
страту – зернової барди.
Вміст сірководню в біогазі (рис. 5) мав значні коливання,
що спостерігається в багатьох біогазових установках
[20], як промислових, так і експериментальних. Це пояс-
нюється, найімовірніше, високою чутливістю сульфат-
редукуючих мікроорганізмів до коливань температури,
складу субстрату та інших умов процесу ферментації,
включно з гідродинамічними. Загалом, зважаючи на ви-
моги до якості біогазу для використання в когенерацій-
них установках (до 100 ррmv), у промисловій установці
потрібно передбачити заходи та обладнання для до
очищення біогазу від сірководню.
Змішану пробу дигестату за останніх п’ять діб експери-
менту піддали детальному аналізу, результати якого
наведені в табл. 4.

Дигестат за складом і концентрацією компонентів
(табл. 4) не може бути використаний для скиду у відкри-
ті водойми, він має підлягати аеробному очищенню в
біофільтрах та/або біологічних ставках. Промислова біо-
газова установка має відповідне обладнання і споруди
для безпечної утилізації дигестату.
Висновки
За результатами експериментальної ферментації мож-
на зробити такі висновки.
Вихід біогазу. У табл. 5 наведений питомий вихід біо-
газу з барди, розрахований за період 13…25 діб фермен-
тації.

Вміст метану в біогазі в період стабільної ферментації
(13…25 діб) становив 60,7 ± 0,8 % об’єму. Це дає змогу
використовувати отримані дані для розрахунків еко-
номічних аспектів роботи промислової біогазової уста-
новки.
Вміст сірководню занадто високий для використання
біогазу в когенераційних установках без попереднього
очищення.
Склад дигестату. Співвідношення С/N у дигестаті стано-
вить 2:1 порівняно з таким у барді (≈14:1). Використання
для промислової біогазової ферментації додаткового
субстрату, лігноцелюлозної біомаси, для оптимального
співвідношення С/N, дасть змогу підвищити вихід біо-
газу.
Навантаження органічною речовиною (OL). Для експе-
рименту було прийняте навантаження реактора ор-
ганічною речовиною ≈4 кг/м3 на добу. Судячи зі співвід-
ношення FOS/TAС, таке навантаження є низьким. Під
час введення в дію та експлуатації промислового реак-
тора для ферментації барди навантаження органічною
речовиною може бути збільшене до 5…6 кг/м3 на добу.
У подальшому в разі використання лігноцелюлозного
косубстрату OL можна буде піднімати до прийнятної
величини.
Отримані дані будуть використані під час введення в
експлуатацію реакторів промислової біогазової уста-
новки для перероблення барди в кількості
1000…1100 м3 на добу.

MODELING THE INITIAL STAGE OF GRAIN STILLAGE BIOGAS FERMENTATION
IN A LABORATORY REACTOR

Abstract. Bioethanol plants face challenges in the maintenance
of using production waste, such as stillage. For every 1 liter of
bioethanol, 10-15 liters of stillage are generated. It can be used
as a raw material for the production of concentrated feed, or
stillage, which is subjected to methane fermentation to obtain
a gaseous energy carrier - biogas. In this way, this Thus, the pro-
duction waste is valorized, and there is a the possibility of re-
placing fossil fuels in the production of bioethanol appears. The commissioning of industrial biogas reactors is
associated with certain difficulties. Our task was to generate a model simulate the commissioning of a reactor
start-up for biogas fermentation of stillage, which comes from an operating plant with a capacity of 170 m3 of
bioethanol per day. To carry out the biogas fermentation process, a laboratory reactor with a working volume of
300 l was used, equipped with a temperature stabilization system (electric heating), an electric mixer, devices for
periodic loading of the substrate and removal of the fermented liquid, a gas removal and accounting system using
a meter. Methane fermentation was controlled by FOS/TAC indicators. The substrate (stillage) was taken directly
from the plant, with a stable technological mode of bioethanol production throughout the experiments. The inoc-
ulum was digestate (a by-product of biogas production) collected from an operating industrial biogas plant. During
the work, the composition of the raw material and inoculum was studied, and the pH and temperature of the
biogas process were controlled. The methanogenesis process took place under thermophilic conditions, at 45-50
°C, the substrate load on dry matter was 4 ± 0.4 kg/m3 per day. The specific biogas yield during the experiment
was 48.5±1.0 m3 per m3 of bard. The methane content in biogas during the period of stable fermentation (13…25
days) was 60.7 ± 0.8% by volume. Based on the results of the experiments, it can be concluded that the addition
of lignocellulosic substrate will increase the biogas yield. Given the level of FOS/TAC, when commissioning and
operating an industrial reactor for wort fermentation, the organic matter loading can be recommended to 5…6
kg/m3 per day. The data obtained will be used when commissioning reactors of an industrial biogas plant for
stillage processing in an amount of 1000…1100 m3 per day.

Overview, issues
Bioethanol is produced in significant quantities from
starch-containing feedstocks—corn, barley, rye, and wheat
[1, 2]. First of all, these are substandard and defective corn
and other cereal grains, grain processing waste that is un-
suitable for food or feed purposes. The raw materials un-
dergo thermoenzymatic treatment, and the resulting sugar
solution is fermented by yeast into ethanol [3].
The by-product after separating ethanol from the culture
fluid is called “post-alcohol distiller's grains,” hereinafter
referred to as “stillage.” Stillage contains all feedstock
components, except for starch and a small proportion of
other carbohydrates that have been converted into ethanol
by yeast. If the quality of the starchy feedstock is satisfac-
tory, the stillage can be used as a raw material for the pro-
duction of feed products [4, 5].
There is another way to utilize stillage—its anaerobic fer-
mentation to produce biogas [6-8]. Recently, more and
more bioethanol enterprises have been using cheap waste
and defective starch-containing raw materials that do not
meet veterinary requirements and may contain toxins,
which makes it impossible to use stillage for feed produc-
tion [9]. In this case, it is advisable to subject stillage to me-
thane fermentation to obtain a gaseous energy source – bi-
ogas or biomethane [10]. This allows for the valorization of
hazardous production waste and the partial replacement of
fossil fuels in bioethanol production [11].
1. Problem statement
As a substrate for anaerobic digestion (AD), stillage has cer-
tain characteristics that must be considered when design-
ing and commissioning industrial biogas plants. The main
differences from traditional substrates for AD, such as

livestock waste and lignocellulosic biomass, are the rela-
tively low carbon-to-nitrogen (C/N) ratio and lower pH
value of stillage. It can be subjected to AD at a fairly high
organic loading (OL) - up to 10 kg per cubic meter of reactor
per day- and hydraulic retention time (HRT) of 6 to 40 days
without washing out the methane-producing microflora.
However, at high loads, the specific amount of biogas pro-
duced (Specific Biogas Production - SBP) decreases from
650 to 550 L kg−1 of organic matter capable of bioconver-
sion (Volatile Solids - VS) [12].
In addition, there are relatively few natural polymer com-
pounds (e.g., cellulose) in stillage. These characteristics ob-
viously influence the microbiological processes in stillage
AD, leading to the formation of biogas. Due to the fact that
the carbon-to-nitrogen (C/N) ratio is too low for biogas fer-
mentation, and the high content of potassium, sulphates,
and phenolic compounds, co-substrates often need to be
added to distiller's grains. The problem can be solved by
adding lignocellulosic biomass. Various agro-industrial
wastes containing cellulose and other carbohydrates are
suitable for this purpose [13, 14].
It is known that the commissioning of biogas reactors is as-
sociated with certain risks. The start of the AD process can
be quite lengthy due to unfavourable changes in pH as a
result of uncontrolled growth of volatile fatty acids (VFAs)
or ammonium compounds concentration, or the negative
influence of sulphur compounds, etc. Excessive accumula-
tion of VFA can cause an unacceptable pH decrease. Con-
versely, a relatively high content of ammonium compounds
in the reactor can lead to exceeding the optimal pH values
for methanogenesis, especially in thermophilic mode.
The enterprise, which produces 170 m3 of bioethanol per
day from starchy feedstocks, uses Very High Gravity (VHG)
fermentation technology [15]. Another feature of the tech-
nology, which results in an atypical stillage composition, is
the use of combined feedstocks—grain sorghum and
starch-containing grain processing waste—which allows for
the production of ethanol mash with a higher concentra-
tion of more than 17% v/v [16]. At the same time, the stil-
lage also has an increased concentration of dry matter (DM)
>12% w/v and VFA (mainly acetic acid – up to 0.5% v/v).
There are plans to build a biogas plant for the utilization of
distiller's grains with the production of biogas.
The aim of the work was to determine the conditions under
which it is possible to carry out the initial stage of the pro-
cess and bring the anaerobic reactor into the operating
thermophilic mode when using a monosubstrate – high-
density stillage.
The tasks were as follows: (1) to generate a model of an
AD reactor start-up for stillage under thermophilic condi-
tions; (2) to determine the period of the reactor's transition
to a stable operating mode at a substrate load of 4 ± 0.4 kg
of DM/m3 per day; (3) to obtain indicators and limits of
their changes, according to which the process will be con-
trolled; (4) to evaluate the specific yield and quality of bio-
gas; (5) to assess the prospect of increasing the reactor load
in terms of DM; (6) to determine the change in the car-
bon/nitrogen ratio in the AD process and the possibility of
increasing reactor productivity by using additional sub-
strates with a high C/N ratio.
2. Materials and methods
For the biogas fermentation process, a laboratory Continu-
ous Stirred Tank Reactor (CSTR) with a working volume of
300 liters was used, equipped with a temperature stabiliza-
tion system (electric heating controlled by the ambient
temperature), an electric stirrer, devices for periodic load-
ing of the substrate and removal of the fermented liquid,
and a gas removal and metering system using a GBR400
meter (the diagram and photo of the reactor are shown in
Fig. 1).

The gas composition (CH4; CO2; H2S) was determined using
a Landtec GEM-500 automatic portable digital gas analyzer
in laboratory tests and using an Aglient 7890B GC System
gas chromatograph (USA). Substrate and culture fluids
analyses were performed in the bioethanol plant labora-
tory using appropriate standardized methods.
Methane fermentation was monitored using FOS/TAC indi-
cators. The FOS (German: Flüchtigen Organischen Säuren)
indicator is a quantitative measure of volatile organic acids,
and TAC (English: Total Anorganic Carbon) is a measure of
alkaline buffer capacity [17, 18].
In addition, for a stable biogas fermentation process, it was
very important to record the temperature in both the reac-
tor and the input raw materials, the DM content, especially
organic dry matter (ODM), and the pH of the reactor media
throughout the anaerobic fermentation process.
The substrate for biogas fermentation (stillage) was taken
directly from the plant during stable bioethanol production
throughout the experiment period. In the technological
process of bioethanol production, sulfuric acid is tradition-
ally used to stabilize the pH of the fermentation medium.
To prevent possible excessive hydrogen sulfide formation
during methane fermentation, sulfuric acid was replaced
with lactic acid in the technological process of bioethanol
production for one week. The table shows the results of the
analysis of the composition of distiller's grains when using
sulfuric and lactic acids. For the analysis, average daily sam-
ples were taken using sulfuric and lactic acids under condi-
tions that prevented mixing samples with different acids.
Measurement methods – in accordance with the State
Standards of Ukraine (DSTU) and Measurement Methods
(MBB 18/01-2019).
2. Analysis of raw materials
The most important indicators of the stillage used in the ex-
periments are shown in Table 1.

Based on the data, the sulphur and other components in
the stillage samples treated with different acids didn't
change much, remaining close to the expanded uncer-
tainty. So, in the AD experiments, we decided not to worry
about these changes in the substrate composition. The ex-
periments were carried out with substrate (stillage) sup-
plied by an operating enterprise, regardless of the acid used
in the technological process.
Before starting the experiments, the laboratory reactor was
loaded with an inoculum, namely digestate (a by-product
of biogas production) selected from an operating industrial
biogas plant. The physicochemical parameters of the inoc-
ulum are shown in Table 2.

4. Description of the experiment
The inoculum for starting fermentation was selected from an
industrial biogas plant, in which a complex substrate with an
unstable ratio of components—lignocellulosic biomass
waste, cattle manure, beet pulp, etc.—was processed in
mesophilic mode (32–36 °C). To prevent thermal stress on
the mesophilic microflora of the inoculum, the AD process in
the laboratory reactor was started in mesophilic mode.
The digestate from the above-mentioned operating biogas
plant (inoculum), delivered for the experiment, had a tem-
perature of about 24 °C. The laboratory reactor was filled
with 300 liters of inoculum and the temperature of the liq-
uid in the reactor was brought to 27.3 °C. After that, 10
liters of stillage were loaded, and the temperature was sta-
bilized at 35±0.5 °C. Further loading was also carried out at
a rate of 10 liters of distiller's grains once a day, based on
an estimated 4 g of organic dry matter per dm3 of reactor
volume and a liquid residence time of 30 days. During load-
ing, the appropriate volume of digestate was withdrawn to
maintain a constant volume of liquid in the reactor.
The temperature in the laboratory reactor was raised by
approximately 2 °C every 24 hours until it reached 40 °C,
then by 1 °C every 24 hours until it reached 45 °C, and by
0.5 °C until it reached ≈50 °C. Such model displays the pro-
cess of an industrial biogas plant start-up when bringing it
into thermophilic mode.
Results and discussion
The data from analyses and measurements throughout the
entire experiment are summarized in Table 3.

The reactor was brought to the planned temperature level
during a period of 14 days (Table 1). Throughout this pe-
riod, the microorganisms adapted to the increase in tem-
perature and substrate load. At the same time, the content
of volatile acids (FOS) increased, and the medium could
have exceeded the optimal pH range of 7–8. In this case, it
would have been necessary to reduce the rate of tempera-
ture rise and/or substrate loading due to exceeding the
adaptive capacity of the microflora in the reactor.
At the period of microorganisms` adaptation, special atten-
tion was paid to changes in the FOS/TAC ratio in the reactor,
ensuring that the content of volatile acids (FOS) relative to
the alkalinity (TAC) of the medium did not exceed the per-
missible value of 0.5. Therefore, the frequency of substrate
loading and the quantity of substrate was not changed.
From the 15th to the 27th day, the metagenesis process
reached a stable state. The alkalinity (TAC) of the inoculum
and substrate remained stable throughout the experiment
due to the constant carbonate content in the distiller's
grains. The relatively low content of ammonium nitrogen in
the distiller's grains did not cause an excessive increase of
pH in the reactor, comparing to processing substrates con-
taining chicken manure [19].
After the adaptation period, the FOS/TAC indicator re-
mained stable at 0.3–0.4.
The inoculum had an initial concentration of 1.3% DM, so
during the adaptation period, this indicator increased until
a balance was established between the substance entering
the reactor and its output with biogas. Fluctuations in the
DM content in the reactor after the process stabilized are
explained by the presence of sediment and its entry into
the liquid samples. The biogas yield during the experiment
is shown in Fig. 3. In general, the dynamics of biogas for-
mation and changes in its composition (Fig. 4) were within
the expected limits, given the stable composition of the
substrate – stillage.
The hydrogen sulphide content in biogas (Fig. 5) fluctuated
significantly, which is also observed in many biogas plants
[20], both industrial and experimental. This is most likely
due to the high sensitivity of sulphate-reducing microor-
ganisms to fluctuations in temperature, substrate composi-
tion, and other fermentation conditions, including hydro-
dynamic ones. In general, given the quality requirements
for biogas for use in cogeneration units (up to 100 ppm),
industrial plants need to provide measures and equipment
for the purification of biogas from hydrogen sulphide.
A mixed sample of digestate from the last five days of the
experiment was subjected to detailed analysis, the results
of which are shown in Table 4:

Due to its composition and concentration of components
(Table 4), digestate cannot be discharged into open water
bodies, but should undergo aerobic treatment in biofilters
and/or biological ponds. An industrial biogas plant has the
appropriate equipment and facilities for the safe disposal
of digestate.
Conclusions
Based on the results of experimental fermentation, the fol-
lowing conclusions can be made:
Biogas yield. Table 5 shows the specific biogas yield from
distiller's grains, calculated for a fermentation period of
13–25 days.

The methane content in biogas during stable fermentation
(13–25 days) was 60.7 ± 0.8% by volume. This allows the
data obtained to be used for calculating the economic as-
pects of industrial biogas plant operation.
The hydrogen sulphide content is too high for the use of
biogas in cogeneration units without prior purification.
Digestate composition. The C/N ratio in the digestate is 2:1
compared to that in distiller's grains (≈14:1). The use of an
additional substrate, lignocellulosic biomass, for industrial
biogas fermentation to achieve an optimal C/N ratio will in-
crease biogas yield.
Organic load (OL). For the experiment, a reactor load of or-
ganic matter of ≈4 kg/m3 per day was adopted. Judging by
the FOS/TAC ratio, this loading is low. When commissioning
and operating an industrial reactor for distiller's grains fer-
mentation, the organic matter loading can be increased to
5–6 kg/m3 per day. In the future, when using lignocellulosic
co-substrate, OL can be increased to an acceptable value.
The data obtained will be used when commissioning reac-
tors of an industrial biogas plant for processing of stillage
at a rate of 1000–1100 m3 per day.

Bioethanol plants face challenges in the maintenance of using production waste, such as stillage. For every 1 liter of bioethanol, 10-15 liters of stillage are generated. It can be used as a raw material for the production of concentrated feed, or stillage, which is subjected to methane fermentation to obtain a gaseous energy carrier - biogas. In this way, this Thus, the production waste is valorized, and there is a the possibility of replacing fossil fuels in the production of bioethanol appears. The commissioning of industrial biogas reactors is associated with certain difficulties. Our task was to generate a model simulate the commissioning of a reactor start-up for biogas fermentation of stillage, which comes from an operating plant with a capacity of 170 m3 of bioethanol per day. To carry out the biogas fermentation process, a laboratory reactor with a working volume of 300 l was used, equipped with a temperature stabilization system (electric heating), an electric mixer, devices for periodic loading of the substrate and removal of the fermented liquid, a gas removal and accounting system using a meter. Methane fermentation was controlled by FOS/TAC indicators. The substrate (stillage) was taken directly from the plant, with a stable technological mode of bioethanol production throughout the experiments. The inoculum was digestate (a by-product of biogas production) collected from an operating industrial biogas plant. During the work, the composition of the raw material and inoculum was studied, and the pH and temperature of the biogas process were controlled. The methanogenesis process took place under thermophilic conditions, at 45-50 °C, the substrate load on dry matter was 4 ± 0.4 kg/m3 per day. The specific biogas yield during the experiment was 48.5±1.0 m3 per m3 of bard. The methane content in biogas during the period of stable fermentation (13…25 days) was 60.7 ± 0.8% by volume. Based on the results of the experiments, it can be concluded that the addition of lignocellulosic substrate will increase the biogas yield. Given the level of FOS/TAC, when commissioning and operating an industrial reactor for wort fermentation, the organic matter loading can be recommended to 5…6 kg/m3 per day. На біоетанольних заводах існує проблема з утилізацією такого відходу виробництва, як барда. На кожний 1 л біоетанолу утворюється 10–5 л барди. Її можна використовувати як сировину для виробництва концентрованих кормів або піддавати метановій ферментації для отримання газоподібного енергоносія – біогазу. У такий спосіб відбувається валоризація цього відходу виробництва, і є можливість заміщення викопних енергоносіїв для виробництва біоетанолу. Введення в дію промислових біогазових реакторів пов’язане з певними складностями. Нашою задачею було моделювання введення в дію реактора для біогазової ферментації барди, що виходить з діючого заводу потужністю 170 м3 біоетанолу на добу. Для проведення процесу біогазової ферментації використовували лабораторний реактор, робочим об’ємом 300 л, обладнаний системою стабілізації температури (електричний підігрів), мішалкою з електроприводом, пристосуваннями для періодичного завантаження субстрату й виводу збродженої рідини, системою виведення й обліку газу за допомогою лічильника. Контроль метанової ферментації здійснювали за показниками FOS/TAC. Субстрат (барда) відбирався безпосередньо з заводу за стабільного технологічного режиму виробництва біоетанолу протягом періоду виконання експериментів. Як інокулят слугував дигестат (побічна речовина після отримання біогазу), який був відібраний з діючої промислової біогазової установки. У процесі роботи досліджувався склад сировини та інокуляту, контролювалися рН та температура біогазового процесу. Процес метаногенезу відбувався у термофільних умовах, при 45–50 оС, навантаження субстратом за сухою речовиною 4 ± 0,4 кг/м3 на добу. Питомий вихід біогазу під час експерименту становив 48,5 ± 1,0 м3 на кожен кубічний метр барди. Вміст метану в біогазі в період стабільної ферментації (13…25 діб) становив 60,7 ± 0,8 % об’єму. Виходячи з результатів експериментів, можна зробити висновок, що додавання лігноцелюлозного субстрату підвищить вихід біогазу. З огляду на рівень FOS/TAС під час введення в дію і експлуатації промислового реактора для ферментації барди навантаження органічною речовиною можна рекомендувати до 5…6 кг/м3 на добу. Отримані дані будуть використані при введенні в експлуатацію реакторів промислової біогазової установки для перероблення барди в кількості 45…55 м3 на добу.
